A 3 enzyme pathway coding for fungal bioluminescence, codon optimized for Nicotiana benthamiana in MoClo-compatible high-copy ampicillin resistance backbones. 

One of the most requested and inspiring biotech projects has been to make glowing plants. Recently, a group out of Moscow has successfully created a glowing plant that is visible with the naked eye. They were able to do this by using the fungal bioluminescence pathway instead of the bacterial bioluminescence pathway, which increased the efficiency of light production in these eukaryotic organisms.

This distribution comprises of {13} genetic parts. {7} of these parts are Luz and Luz alternatives, the main light producing enzyme. {3} of these parts are 2 enzymes that complete the biosynthetic pathway necessary to sustain light production. 1 enzyme, HispS, has been split into 2 parts because it was too large for synthesis directly. It can be put back together with BglII or BsaI with the overhang sequence of GATC. The remaining {3} parts are vector components to put test these genes quickly while a FreeGenes Plant toolkit is put together.      

Shipped as purified and dried down plasmid DNA stained with Cresol Red in one 96-well plate. Each well contains approximately 50 ng of DNA. Cresol Red will not impact plasmid transformation. The plate map can be found here.

Full history of modifications are available in the snapgene files in sequence_files.zip. Genbank files are also provided in genbank_files.zip. The exact sequences ordered from Twist are available in glowing_plant_order.csv. The genes are flanked by experimentally validated orthogonal primer pairs, and a I-SceI barcode. Full set of primer pairs for all FreeGenes parts are in a git repo. Detailed information on changes can be found in the curators_notes.md.

Note: Partial plasmid duplication has been found for BBF10K_003492 (H3H test vector). This partial duplication should not impact transformation, outgrowth or primer attachment.
